Novel fusion peptide?mediated siRNA delivery using self?assembled nanocomplex

Abstract Background Gene silencing using siRNA can be a new potent strategy to treat many incurable diseases at the genetic level, including cancer and viral infections. Treatments essentially requires an efficient safe method of delivering into cells while maintaining its stability. Thus, we designed novel synergistic fusion peptides, i.e., SPACE oligoarginine. Results Among peptides siRNAs, nanocomplexes have enhanced cellular uptake gene effect in vitro improved retention effects siRNAs vivo. Oligoarginine could attract electrostatically form stable self-assembled nanocomplexes, peptide interact with membrane via hydrogen bonding. Therefore, showed evident glyceraldehyde 3-phosphate dehydrogenase (GAPDH) lipid raft-mediated endocytosis pathway, especially HDFn skin, all were biocompatible. Also, intratumorally injected had increased time site tumor. Finally, demonstrated significant vivo without overt tissue damage immune cell infiltration. Conclusions The nanocomplex become platform for delivery tissues various target through silencing.
